KPV Overview
KPV is a naturally occurring tripeptide composed of lysine, proline, and valine. It represents the C-terminal three-amino-acid sequence of alpha-melanocyte-stimulating hormone (α-MSH), a peptide involved in numerous inflammatory and immune-regulatory processes. Despite its small size, KPV retains significant biological activity associated with the anti-inflammatory properties of the parent α-MSH peptide. Experimental studies have examined KPV in intestinal epithelial cells, immune-related models, and animal models of inflammation. Research has particularly focused on inflammatory signaling pathways including NF-κB and MAPK, as well as cytokine signaling, cellular migration, and intestinal inflammatory responses. Evidence for KPV remains predominantly preclinical, and the compound should be presented as an investigational research material rather than an established therapeutic agent.
History
Scientific interest in KPV developed from research into the biological activity of α-MSH and its peptide fragments. By 1990, published experiments were demonstrating anti-inflammatory activity from α-MSH(11–13), the C-terminal tripeptide now commonly identified as KPV. Research subsequently focused on determining whether the shortened KPV sequence could retain the anti-inflammatory properties of full-length α-MSH without requiring the complete melanocortin peptide structure. Later studies found significant activity in preclinical models of intestinal inflammation and investigated mechanisms independent of classical melanocortin-receptor signaling. In 2008, research involving intestinal epithelial and immune-cell models demonstrated PepT1-mediated transport of KPV and reported reduced activation of NF-κB and MAPK inflammatory signaling following intracellular uptake. These findings helped establish intestinal inflammation and epithelial signaling as major areas of contemporary KPV research.
